En campagne pour sa réélection en vue de l'élection présidentielle de 1964, il circule dans Dallas le 22 novembre 1963 à bord d'un véhicule découvert devant un public nombreux : alors qu'il traverse Dealey Plaza, des coups de feu retentissent et des tirs l'atteignent mortellement.
